Identification of the transcriptional regulatory sequences of human calponin promoter and their use in targeting conditionally replicating herpes vector to malignant human soft tissue and bone tumors. We created transcriptionally targeted oncolytic herpes simplex virus vector bM24TE in which replication is driven by the TE promoter. The WntbetacateninTcell factor Tcf pathway is aberrantly upregulated in the majority of colorectal cancers CRC and hepatoblastomas due to either an APC or betacatenin gene mutation. Intratumoral injection of bM24TE significantly reduced the growth of betacatenininactive A549 tumors. This vector efficiently and specifically replicated in and killed tumor cells with strong betacateninTcf signaling.

Overexpression of Icat induces G2 arrest and cell death in tumor cell mutants for adenomatous polyposis coli, betacatenin, or Axin. Fusion of the BCL9 HD2 domain to E1A increases the cytopathic effect of an oncolytic adenovirus that targets colon cancer cells.
